Evidence that the transcriptional regulators SIN 3 and RPD 3 , and a novel gene ( SDS 3 ) with similar functions , are involved in transcriptional silencing in S . cerevisiae . ^^^ SDS 16 and SDS 6 are known transcriptional modifiers , SIN 3 ( RPD1 / UME4 / SDI1 / GAM2 ) and RPD 3 ( SDI 2 ) , respectively , while the third is a novel gene , SDS 3 . ^^^ SDS 3 shares the meiotic functions of SIN 3 and RPD 3 in that it represses IME 2 in haploid cells and is necessary for sporulation in diploid cells . ^^^ However , sds 3 mutations differ from sin 3 and rpd 3 mutations in that they do not derepress TRK 2 . ^^^ Epistasis studies suggest that SDS 3 works in a different pathway from RPD 3 and SIN 3 to affect silencing at HMR . ^^^

The remaining gene from this screen described in this report is SDS 3 , previously identified in a screen for mutations that increase silencing at HML , HMR , and telomere linked genes , a phenotype also seen in sin 3 and rpd 3 mutants . ^^^ Genetic analysis demonstrates that SDS 3 functions in the same genetic pathway as SIN 3 and RPD 3 , and coimmunoprecipitation experiments show that Sds 3 is physically present in the Sin 3 complex . . ^^^

Sds 3 ( suppressor of defective silencing 3 ) is an integral component of the yeast Sin 3 [ middle dot ] Rpd 3 histone deacetylase complex and is required for histone deacetylase activity . ^^^ In addition , sds 3 mutants have phenotypes very similar to those seen in sin 3 and rpd 3 mutants , suggesting that it functions in the same genetic pathway . ^^^
