MDM 2 HDAC 1 mediated deacetylation of p 53 is required for its degradation . ^^^ Here we show that MDM 2 can promote p 53 deacetylation by recruiting a complex containing HDAC 1 . ^^^ The HDAC 1 complex binds MDM 2 in a p 53 independent manner and deacetylates p 53 at all known acetylated lysines in vivo . ^^^ Ectopic expression of a dominant negative HDAC 1 mutant restores p 53 acetylation in the presence of MDM 2 , whereas wild type HDAC 1 and MDM 2 deacetylate p 53 synergistically . ^^^ Fibroblasts overexpressing a dominant negative HDAC 1 mutant display enhanced DNA damage induced p 53 acetylation , increased levels of p 53 and a more pronounced induction of p 21 and MDM 2 . ^^^ |
