A human SWI2 / SNF2 homologue , hBrm , is a positive participant in glucocorticoid receptor mediated transcription , but its mechanism of action is not known . ^^^ These results highlight a new role for RB , which is to interact with hBrm in order to potentiate glucocorticoid receptor activated transcription . . ^^^

In cells failing to express the endogenous gene , transfected hbrm cooperates with the glucocorticoid receptor ( GR ) in transcriptional activation . ^^^ The co operation between hbrm and GR required the DNA binding domain of GR and two separated regions of the hbrm protein , including a domain with homology to known helicases . . ^^^

The cell cycle regulator , retinoblastoma protein , is known to potentiate glucocorticoid receptor activated transcription through the interaction of its pocket domain with the transcription coactivator , hBRM . ^^^ We now show that glucocorticoid receptor induced apoptosis is also dependent on both the retinoblastoma protein and hBRM . p 107 and p 130 , which share extensive sequence homology with the pocket domain of the retinoblastoma protein but not its N terminal region , also interact with hBRM but do not support either glucocorticoid receptor dependent activity . ^^^ This effect probably results from the promotion of glucocorticoid receptor targeted chromatin remodeling by the hBRM containing SWI / SNF complex because the N terminal domain of the retinoblastoma protein enhances glucocorticoid receptor hBRM interactions . ^^^ These results highlight that , besides the interaction between hBRM and the pocket domain of RB , the N terminal region of the retinoblastoma protein is also essential for glucocorticoid receptor induced apoptosis and the potentiation of glucocorticoid receptor mediated transcription and provide a basis for functional distinction between the retinoblastoma protein and its homologues . . ^^^

The DNA dependent ATPase domains of brm and SWI 2 , but not ISWI , are functionally interchangeable ; a chimeric SWI 2 brm protein partially rescued the slow growth of swi 2 cells and supported transcriptional activation mediated by the glucocorticoid receptor in vivo in yeast cells . ^^^

The SWI 1 , SWI 2 , and SWI 3 proteins , which are required for regulated transcription of numerous yeast genes , were found also to be essential for rat glucocorticoid receptor function in yeast ; the receptor failed to activate transcription in strains with mutations in the SWI 1 , SWI 2 , or SWI 3 genes . ^^^ Immunoprecipitation of glucocorticoid receptor derivatives from wild type ( SWI+ ) yeast extracts coprecipitated the SWI 3 protein ; such receptor SWI 3 complexes were not detected in swi 1 or swi 2 mutant strains , implying that a complex of multiple SWI proteins may associate with the receptor . ^^^

It was recently reported that the mammalian glucocorticoid receptor failed to activate transcription when transiently expressed in swi 1 , swi 2 or swi 3 yeast strains . ^^^
