We investigated overexpression of cyclin D 1 ( CCND 1 ) , cyclin D 2 ( CCND 2 ) , cyclin E ( CCNE ) , CDK 2 , and CDK 4 , in addition to p 27 , in 260 gastric cancer cases on the basis of Western blots , reverse transcriptase polymerase chain reaction Southern blots , and immunohistochemistry to clarify the roles of these proteins in tumor progression and prognosis . ^^^ Immunohistochemistry demonstrated 21 . 5 % , 34 . 2 % , 30 . 4 % , 44 . 2 % , and 48 . 0 % positivity for CCND 1 , CCND 2 , CCNE , CDK 2 , and CDK 4 , respectively , in the 260 gastric cancer cases . ^^^ Overexpression of CCND 2 and CDK 4 significantly correlated with tumor progression . ^^^ Survival curves showed CCND 2 ( especially cytoplasmic staining ) and CDK 4 positivity to be associated with a poor prognosis and CCNE positivity with a better prognosis . ^^^ There were no direct links among CCND 2 , CCNE , CDK 4 , and p 27 . ^^^ |

To compare the expression status of G 1 cyclins , these EBV associated lymphoma lines ( 6 EBV [ ] human SCID mouse lymphoma lines , 13 human B cell lymphomas and 8 samples of human tonsil tissue ) were examined by reverse transcription polymerase chain reaction Southern blotting , Western blotting and immunohistochemistry . mRNA expression of cyclin D 1 ( CCND 1 ) , cyclin D 2 ( CCND 2 ) , cyclin E ( CCNE ) , cyclin dependent kinase 2 ( CDK 2 ) and 4 ( CDK 4 ) was found in all 3 types of lymphomas . ^^^ Immunoprecipitation of EBV ( + ) and ( ) SCID mouse lymphomas for CCND 1 , CCND 2 and CCNE vs . p 21 , PCNA and CDK 2 or CDK 4 demonstrated that , in EBV ( + ) SCID lines , CCND2 / CDK4 complexes were present without binding to p 21 , suggesting independence from p 21 regulation . ^^^ |

The expression status of the three cyclin D genes ( CCND 1 , CCND 2 and CCND 3 ) , the two cyclin D dependent kinase genes ( CDK 4 and CDK 6 ) and the p 16 ( INK4a ) gene was studied in a series of 47 Wilms ' tumors , 16 normal mature kidneys and two fetal kidneys . ^^^ We showed predominant overexpression of CCND 2 and CDK 4 compared to CCND1 / D3 and CDK 6 respectively . ^^^ This suggests that dysregulation of CCND 2 and CDK 4 plays a specific role in WT tumorigenesis . . ^^^ |
