To investigate whether alteration of other components in pRb pathway is an alternative mechanism underlying the development of these chemically induced lymphomas , we have examined the genetic status of Rb 1 , Ccnd 1 and Cdk 4 genes that encode retinoblastoma protein , cyclin D 1 and cyclin dependence kinase 4 , respectively . ^^^ Gross alterations of the Rb 1 , Ccnd 1 , and Cdk 4 genes were not detected by Southern analysis in any of the tumors examined . ^^^ These results indicate that the mechanisms underlying the development of 2 ' , 3 ' dideoxycytidine and 1 , 3 butadiene induced lymphomas involve inactivation of p16 / p15 cyclin dependent kinase inhibitors but not genomic alterations of the Rb 1 , Ccnd 1 and Cdk 4 genes . . ^^^ |

To compare the expression status of G 1 cyclins , these EBV associated lymphoma lines ( 6 EBV [ ] human SCID mouse lymphoma lines , 13 human B cell lymphomas and 8 samples of human tonsil tissue ) were examined by reverse transcription polymerase chain reaction Southern blotting , Western blotting and immunohistochemistry . mRNA expression of cyclin D 1 ( CCND 1 ) , cyclin D 2 ( CCND 2 ) , cyclin E ( CCNE ) , cyclin dependent kinase 2 ( CDK 2 ) and 4 ( CDK 4 ) was found in all 3 types of lymphomas . ^^^ Immunoprecipitation of EBV ( + ) and ( ) SCID mouse lymphomas for CCND 1 , CCND 2 and CCNE vs . p 21 , PCNA and CDK 2 or CDK 4 demonstrated that , in EBV ( + ) SCID lines , CCND2 / CDK4 complexes were present without binding to p 21 , suggesting independence from p 21 regulation . ^^^ |
