Gankyrin is an ankyrin repeat oncoprotein that interacts with CDK 4 kinase and the S 6 ATPase of the 26 S proteasome . ^^^ Overexpression and precipitation of tagged gankyrin from cultured cells detects a complex containing co transfected tagged S 6 ATPase ( or endogenous S 6 ) and endogenous cyclin D dependent kinase CDK 4 . ^^^ The proteasomal ATPases are part of the AAA ( ATPases associated with diverse cellular activities ) family , members of which are molecular chaperones ; gankyrin complexes may therefore influence CDK 4 function during oncogenesis . . ^^^ |

Novel insights into the INK 4 CDK4 / 6 Rb pathway : counter action of gankyrin against INK 4 proteins regulates the CDK 4 mediated phosphorylation of Rb . ^^^ We further demonstrated a different mechanism for gankyrin to facilitate the phosphorylation of Rb , by binding with cyclin dependent kinase 4 ( CDK 4 ) . ^^^ We then showed that binding of gankyrin to CDK 4 and the consequent counter action of p 16 function were not affected by the Rb binding peptide 176LHLACDEERN185 , indicating that the two mechanisms are independent . ^^^ While the Rb binding motif is located at the fifth ankyrin repeat , truncation mutants of gankyrin , with the first three or four ankyrin repeats remaining , are sufficient for binding to CDK 4 and for counteracting the inhibitory function of p 16 . ^^^ |

This study was undertaken to observe the effect of signal regulatory proteinalpha 1 ( Sirpalpha 1 ) on gankyrin , cyclin D 1 , CDK 4 and Fas expression in Sk hep 1 mouse hepatoma carcinoma cell line . ^^^ The protein expressions of gankyrin , cyclin D 1 , CDK 4 and Fas in various Sk hep 1 lines were determined by Western blotting . ^^^ Sirpalpha1P4Y2 downregulation of gankyrin expression was greater than that of Sirpalpha 1 ( P < 0 . 05 ) , but no significant effect of Sirpalpha 1 and Sirpalpha1P4Y2 on CDK 4 and Fas protein expression was observed in transfected Sk hep 1 lines ( P > 0 . 05 ) . ^^^ |
