To investigate the mechanism involved , we studied the effects of estrogen on GH signaling through Janus kinase ( JAK ) 2 and the signal transducers and activators of transcription ( STATs ) in HEK 293 cells stably expressing the GH receptor ( 293GHR ) , HuH 7 ( hepatoma ) and T 47D ( breast cancer ) cells . 293GHR cells were transiently transfected with an estrogen receptor alpha expression plasmid and luciferase reporters with binding elements for STAT 3 and STAT 5 or the beta casein promoter . ^^^

The present study investigates the importance of the GHR cytoplasmic domain in the activation of STAT 3 and STAT5b . ^^^ STAT5b and STAT 3 , when each was coexpressed in COS 7 cells with WT GHR and WT JAK 2 , were both strongly tyrosine phosphorylated in response to GH . ^^^ Coexpression of STAT 3 with GHR / JAK 2 chimeras resulted in a strong GH independent tyrosine phosphorylation of STAT 3 that was 40 % as active as that seen with WT GHR plus WT JAK 2 , whereas STAT5b was more minimally phosphorylated ( 13 % of WT GHR plus WT JAK 2 ) when coexpressed with chimeras devoid of the GHR cytoplasmic domain . ^^^ Transient coexpression of each STAT together with WT JAK 2 and GHR COOH terminal truncation mutants indicated that a GH induced STAT 3 DNA binding complex , but not a STAT5b DNA binding complex , was detectable when a GHR devoid of 85 % of the cytoplasmic domain COOH terminus ( but eliciting significant JAK 2 tyrosine phosphorylation ) was expressed . ^^^ We conclude that GH induces activation of STAT 3 and STAT5b by two different pathways : one primarily dependent on activation of JAK 2 ( STAT 3 ) and another that is additionally reliant on the presence of an intact and tyrosine phosphorylated GHR cytoplasmic domain ( STAT5b ) . . ^^^

Our results demonstrate that ( 1 ) both GHR mRNA and GHR immunoreactivity are present in HL 60 cells ; ( 2 ) GH treatment results in an increase in the phosphorylation of the GHR associated Jak 2 and Stat 3 proteins , indicating the ability of the hormone to induce receptor activation ; and ( 3 ) activation of GHR increases the activity of Akt , a serine / threonine kinase that plays a prominent role in the regulation of cell survival . ^^^
